In bacterial disease like Chlamydia & Gonorrhea, in traditional swab test, they look out for bacterial culture, that is, it involves attempting to grow bacteria out of samples that were taken directly from the cervix or urethra.
In Urine test for STD detection, they check for the presence of bacterial DNA using various techniques like LCR (ligase chain reaction) or DNA amplification. These types of testing are sensitive to even very small amounts of bacterial DNA found, even better, they do not require a live bacterial sample.
